DNA is a colorless biomolecule that cannot be seen on the gel during gel electrophoresis. DNA Loading Dye Kits contain bromophenol blue, Xylene Cyanol, and Orange G dyes that are mixed with the DNA sample. The loading dye kits are used for easy loading of DNA on the gel and to keep a visual track of DNA during gel electrophoresis.

The dyes in the kit help the user to determine the maximum resolution of the DNA fragments on the gel. Also, the glycerol or Ficoll present in the DNA loading kit provides an additional density to the DNA molecules which further helps DNA to come out from the well. DNA loading dyes are used during gel electrophoresis for the smooth loading of DNA into wells and for tracking DNA migration across the gel. They are used for both agarose and native polyacrylamide gels.

The composition of the dye can vary but will usually contain some of the following tracking dyes: Bromophenol Blue, Orange G, or Xylene Cyanol FF. Varying concentrations of DNA loading dyes are available, and the concentration can also be adjusted according to experimental needs. EDTA may be included in some dyes as a chelating agent and to inhibit nuclease activity, while glycerol helps the DNA in the ladder and sample form a layer at the bottom of the well.
