Long read sequencing which is also known as third-generation sequencing, allows sequencing of longer deoxyribonucleic acid (DNA) fragments as compared to traditional short read sequencing technologies. There are two main products in the long read sequencing market - PacBio sequencing systems and Oxford nanopore sequencing systems.
PacBio sequencing systems use single-molecule real-time (SMRT) sequencing to read entire DNA molecules. This method detects fluorescence from individual DNA polymerase complexes to determine the sequence in real time as synthesis occurs. The advantage is that it can generate read lengths of 10-20kb on average, which is useful for sequencing complex genomes and determining haplotypes. However, it has a higher error rate of 15% as compared to short read technologies. Oxford nanopore sequencing systems are based on protein nanopores and can read DNA strands passing through these nanopores. It can produce very long reads in the range of 50kb-1Mb. Though the error rate is higher at 6-15%, ongoing developments aim to reduce it. An added advantage is that the portable MinION device allows real-time and off-grid sequencing.
Long read sequencing technologies have enabled sequencing of much longer fragments as compared to short read methods. While error rates are still relatively high, ongoing research promises to reduce it further and allow more accurate assembly of complex genomes.
